Caractérisation moléculaire du microenvironnement cellulaire de lymphomes B de patients immunodéprimés. (Service de Pathologie Clinique CHUV, Prof. L. de Leval)

Porhyrogenis Foundation sustained Mrs Adamantia Chantziou for 18 months in my laboratory at the Institute of Pathology of CHUV. The aim of her research was to characterize the immune landscape of Hodgkin lymphoma (HL) in patients infected or not with the Human Immunodeficiency Virus (HIV), and with respect to Epstein Barr Virus (EBV) infection. To this end, multiplexed imaging using Vectra Polaris (Akoya), and tissue gene expression profiling assays (HTG EdgeSeq and GeoMx Digital Spatial Profiler (DSP)), were developed and applied to a cohort of 29 tissues from HL patients, retrieved from our files and from Attikon University Hospital, Athens, Greece. The analysis of the tumor microenvironment in HL revealed that HIV infection does not impact the neoplastic cell densities (cells/mm²). It was found that HIV infection impacts the innate immunity subsets, by decreasing the cell densities of antigen presentation professional dendritic cells, and by increasing the cell densities of macrophages. The transcriptomic analysis revealed that HIV infection impacts the functions of adaptive immunity T cell subsets, namely in the capacity for antigen presentation and exhaustion.
